Objective: To established a model of regional chronic cerebral hypoperfusion, we used improved line suture of rats induced MCAO model. Then we explored the change of nerve cell morphology and hs-CRP in serum at this special condition, in order to provide a basis for the further study of cerebrovascular disease and retrograde neurological disease.Methods: 1. 60 healthy adult male SD rats were randomly divided into control group, sham-operated group and operated group including four subgroup, that is 1 d, 3 d, 7 d, 14 d post of operation. We used an improved line suture to induce MCAO model. Then we determined the successful establishment of hypoperfusion model by scoring of neural behavior, TTC staining. 2. Then detected and observed the apoptosis expression of nerve cell and the concentration of hs-CRP in serum of rCCH rats at different time points with immunohistochemistry and ELISA. Results: 1. After an operation, the score of neurological behavior in both of the two groups were≤1point, and there was no significant difference between groups(P>0.05).We found no regional brain infarction neither in the control group nor in the experimental group with TTC staining in rats'brain, and could see disintegration of cytoplasm in optic nerve cells with electron microscopy.2. Detection of Hs-CRP levels in serum and neuronal apoptosis:At 1 d, 3 d, and 7 d postoperatively, the hs-CRP content in rCCH rat serum were higher than those of control group, and there was insignificant difference (P>0.05); At 14days postoperatively, the hs-CRP content in rCCH rat serum were higher than those of control group(P<0.05), there was a significant difference (P<0.05). Moreover, the hs-CRP content increasing with the days, which shows hs-CRP level in rCCH rat serum continued to increase along with the experimental time, during the interval 1 and 2 weeks post surgery. In contrast, the apoptotic of nerve cells increasd sharply within 1 day post surgey and astrocyte increased significantly. There was a significant difference when comparing the group of 1d,3days postoperatively and the control group (P<0.01). The apoptotic cells tended to increase as before between 7 days and 12 days postoperatively. However, the number which increased seemed to be decreased more or less owing to the time interval.Conclusions:1,Through improved suture method induced MCAO model, we can successful establish reliable regional chronic cerebral hypoperfusion model in rats2,Regional chronic cerebral hypoperfusion may induces neuronal apoptpsis deucedly.3,Neuronal apoptosis of rCCH rat's MCA territory got serious gradually with the increased time.
