Objective: To compare the effects of bone marrow-derived stem cells (MSC) transplantation and bone marrow cell mobilization by injection of granulocyte colony-stimulating factor treatment the liver damage of rats.Methods: BMSCs isolated from rat bone marrow were purified and proliferated in vitro.The model of a rat liver damage were made by intraperitoneal injection of 40﹪(v / v) carbon tetrachloride solution in vegetable oil, 1ml/kg, injected twice a week for 4 weeks and confirmed by Plasma ALT and AST and pathological observation, then the model were treated with transplant BMSCs and bone marrow cell mobilization respectively. In order to prevent liver damage of rats spontaneous recovery, the rats were continued the carbon tetrachloride injection. In order to tracing the BMSCs transplanted cells, the BMSCs were markered with DAPI and BrdU respectively.Results: The rat BMSCs showed fibroblast-cell-like shape. It was confirmed that the BMSCs of rats were purified by detecting the positive surface antigen CD44 and negative surface antigen CD34. CD44 expression of rBMSCs is positive, but not CD34. After 40﹪CCl4 injection for 4 weeks, the regenerating liver cells and fibers fusion were observed in experimental rats increased, there were the early formation cord of fibrous tissue hyperplasia, but it did not convert Hepatic lobule, and the level of ALT and AST increased. It showed that the liver injury model of rats was successful. After treated with transplant BMSCs and bone marrow cell mobilization for 3 weeks respectively, the general state of health of the model rats had improved, the hair color got to bright, the body weight increased and the level of ALT and AST increased. But both mobilization and transplantation could not completely recover the blood biochemistry to normal level. The labeletd stem cells were observed in the liver tissue of transplantation rat.,it showed that transplanted stem cells home to the liver. The numbers of mono-nuclear cell increased in peripheral blood of mobilized stem cell rat, the results also showed that the mononuclear cells in bone marrow of rats could be mobilized by the granulocyte colony-stimulating factor. The results of blood biochemistry examination also showed that the treatment of BMSCs transplant was better than that of the self-stem cell mobilization . Pathological examination results also showed the similar conclusion.Conclusion: 1. The tissue constructure and function of liver injuried were improved by G-CSF mobilized autologous and transplantation of BMSCs purified and amplified in vitro, and stem cell transplantation is better than G-CSF mobilization. 2. Ordinary injection needle was changed into a transplant needle to increase the successful rate of transplantation by portal vein and reduced the tissue injury. 3. There was not significant difference in labeling BMSCs between Brdu and DAPI methods, but Brdu was more convenient.
