Effect Of ω-3 Pufas On The Retinal Neovascularrization

Object:TO explore the effect ofω-3 PUFAs on the retinal neovascularrization.Methods:Thirty two 7 days old clearing Wistar pups with 4 mother mice were randomly divided into four groups to make Retinopathy of Premature (ROP) model:control group(A),low dose experimental group(B),middle dose experimental group(C) and high dose experimental group(D), each group was with a mother mice, and experimental group was feded by different dose of co-3PUFAs as 2.5mg/kg/d,7.5mg/kg/d and 15mg/kg/d. The numbers of endotheliocyte nuclei of neovascularization which broke through the retinal inner limiting memberane was detcted by immunohistochemistry,the expression of Regulators of G—protein Signaling (RGS5) in retinal was detected by Western blot.Results:The numbers of endotheliocyte nuclei of neovascularization which broke through the retinal inner limiting memberane were(17.00±3.42)%(15.63±2.67) (9.13±1.96),(8.38±2.19) in four groups respecitively. The ration between RGS5 andp-Actin were(0.24±0.96),(0.36±0.08),(0.51±0.23),(0.66±0.19) in four groups respecitively.There was significan difference found between control group and middle dose experimental group(P<0.001),also there were difference for low dose experimental group and high dose experimental group compared with the control group(P<0.05).but there was no statistical differencewan found between middle dose experimental group and high dose experimental group(P>0.05).Conclusion:RGS5 take a part in retinal neovascularrization, The dose ofω-3 PUFAs take effect on RGS5, and had a protective effect against pathological angiogenesis in retinal.
