Effects Of Endostar On Immune State Of Lung Metastasis Of Melanoma B16 Cells In C57bl/6 Mice

Objective: Study on the effects of endostar (rhEndostatin) on the mRNA expression of CCL12, CCL21, Foxp3, TGF-β1 and positive rates of HIF-1α, VEGF protein of lung metastasis of melanoma B16 cells in C57BL/6 mice. And provide a new theoretical support for endostar of clinical use.Methods: The B16 experimental pulmonary metastases was prepared by injecting the B16 cell suspension via lateral tail vein of C57BL/6 mouse. After tumor inoculation, the mice were randomly divide into two groups: one group were administered endostar, 20mg/kg/d, every day for 10 days; the other group were given sodium chloride. The mice were sacrificed 24 hours after the last time of administered. The body weight was measured at 0, 3, 5, 7, 9, 11, 13d, respectively. The number of black metastatic colonies in lungs were counted under a dissecting microscope. Immunohistochemistry was used to detect the expression of HIF-1αprotein and VEGF protein. RT-PCR and real-time PCR were used to detect the expression of CCL12 mRNA, CCL21 mRNA, Foxp3 mRNA and TGF-β1 mRNA of lung tissue.Results:(1) The body weight of the two group were increased, and there is no significant difference. (2)The significant reduction in the number of pulmonary tumors was observed following treatmment of mice with endostar. (3)The positive rates of HIF1α, VEGF in treatment group were lower than in control group (P<0.05). (4) The CCL12 mRNA expression in treatment group was increased compared with in the control group (P<0.05). The CCL21 mRNA, Foxp3 mRNA and TGF-β1 mRNA expression were less in treatment group than in control group (p<0.05).Conclusion: The results suggest that endostar has strong inhibitory effect on the metastasis of lung B16 melanoma in C57BL/6 mouse. And provide a new theoretical support for endostar of clinical use.
