Objective: To investigate the functions of Th17 cells of peripheral blood in allergic rhinitis. Methods: 10 patients of severe perennial allergic rhinitis (AR) due to mite and 10 healthy controls (HC) were enrolled. Peripheral blood mononuclear cells (PBMCs) were collected ,divided into three groups : ionomycin group(blank group) ,adding ionomycin 1ug/ml and monesin 2umol/l;mite group , adding ionomycin 1ug/ml,mite allergen nDer p1 1ug and monesin 2umol/l; PMA group , adding PMA 20ng/ml,ionomycin 1ug/ml and monesin 2umol/l. Cells were activated for 6h at 37°C in a 5% CO 2 atmosphere, Cells were harvested for cell membrane and intracellular staining , then analyzed on flow cytometer. Results: The percent of Th17 cells in CD4+T of each HC group was lower than that of AR. After the stimulation with PMA, the percent of Th17 cells in CD4+T of HC and AR increased (p< 0.05) and the AR increased more.After the stimulation with nDer p1, the percent of Th17 cells in CD4+T of HC had no significantly change,but the the percent of Th17 cells in CD4+T of AR decreased significantly(p<0.05) .Conclusions : This suggested that Th17 cells maybe had an influence of AR. PMA could increase the number of CD4+T cells that could secret IL-17 in the vitro incubation. Results: The percent of memory Th2 cells of HC ionomycin group was higher than that of AR; after the stimulation with nDer p1, the percent of memory Th2 cells of HC had no significantly change, but the percent of memory Th2 cells of AR increased and the TcmTh2 increased significantly(p< 0.05). The percent of memory Th1 cells of HC blank group was higher than that of AR, after the stimulation with nDer p1, they all decreased but the HC decreased more although it had no significance. After the stimulation with PMA, the percent of memory Th2 cells of AR and HC decreased significantly(p< 0.05); the percent of memory Th1 cells of AR and HC increased significantly(p < 0.05). Conclusions: The AR had special memory Th2 cells to mite allergen. PMA could decrease the number of CD4+T cells that could secret IL-4 and increase the number of CD4+T cells that could secret IFN -γ.
