Objective: This study we detect the expressions of STAT3 and VEGF in gastric carcinoma tissues and normal gastric mucosa by immunohis to chemical technique ,analyze the relationship between the expressions and the ability of invasion and metastasis of tumor cells,the relationship between the expressions and the biological behavior of gastric carcinoma and the relationship between the expressions and the prognosis.So we can provide the theoretical foundation for prognosticating the invasion and metastasis of gastric carcinoma,evaluating the prognosis and guiding the reasonable comprehensive treatment.Method: In this study, we select 76 samples of gastric carcinoma , who had taken the radical operation and proved by pathology in the tumor department of surgery of the hospital affiliated HeBei University from June 2003 to Dec 2008. None of them underwent preoperative chemotherapy and/or radiation therapy.35 samples of normal gastric mucosa tissue served as contrast. All gastric carcinoma clinical records registered according to gastric carcinoma treatment criterion. The last follow up date wasJune.30,2006and calculte closure data. In this study, follow up 65 cases and lose 11 cases. The median duration of follow-up was 1268 days. At the last follow-up examination, 42 patients were still alive, whereas 23 patients had died, all died of the recur and metastasis of tumor . The expressions of STAT3 and VEGF in gastric carcinoma tissues and normal gastric mucosa were detected by immunohistochemical technique in this study. The relationship between the expressions of STAT3 and VEGF and the biological behavior of gastric carcinoma were analyzed , including sex,age,tumor location,size,gross type,differentiation degree,depth of invasion,lymph node metastasis and TNM stage etc.Theχ2 test was perfomed to determin the relationship between the expressions of STAT3 and VEGF and the clinical pathology. Survival durations were calculated using the Kaplan-Meier method. All data are analyzed by statistics software SPSS 11.0.Results:1. The expressions of STAT3 and VEGF in gastric carcinoma tissues are significantly higher than those in normal gastric mucosa.It shows the aggressive nature of human gastric carcinoma is related to mutations of various oncogenes and tumor suppressor genes and abnormalities in several growth factors and their receptors . 2. The expressions of STAT3,VEGF in gastric carcinoma tissues show no difference in patients'sex,age,tumor location and size.3. The expressions of STAT3 in gastric carcinoma tissues become significantly higher along with differentiation degree to lower,the depth of invasion to deeper, lymph node metastasis, TNM stage to higher. The expressions of VEGF in gastric carcinoma tissues become significantly higher along with the depth of invasion to deeper, lymph node metastasis, TNM stage to higher and the expressions of VEGF show no difference in differentiation degree. It shows the expressions of STAT3 and VEGF are correlated with clinical pathology progress of gastric carcinoma.Strong STAT3 and VEGF expression indicates the ability of the invasion and metastasis of gastric carcinoma become higher, and the ability of local anti-invasion and anti-metastasis become lower.4. In gastric carcinoma tissues,the relationship between STAT3 expression and survival rate: The medium survival time in 65cases who had a tumor with negative, The impact of VEGF expression on survival duration was not statistically significant, however. It shows abnormal STAT3 expression and activation may be a more powerful predictor of patient outcome. STAT3 is clearly a better independently predictor.5. In gastric carcinoma tissues aberrant STAT3 activation may have directly caused the VEGF over-expression, VEGF expression was highly correlated with STAT3 expression. It shows that STAT3 have central role in regulating multiple genes that lead to aggressive growth and metastasis, and that dysregulated expression of STAT3 may influence many aspects of gastric cancer biology, including VEGF overexpression.Conclusions:STAT3 is an essential transcription factor for many genes that may regulate many aspects of cancer biology, including survival, growth, invasion, and angiogenesis. In this study we discover that the abnomal expression of STAT3 may have directly caused the VEGF overexpression, abnormal STAT3 expression and activation may be a more powerful predictor of patient outcome. Unite examining the expressions of STAT3 and VEGF in gastric carcinoma tissues can be useful in prognosticating the invasion of gastric carcinoma,evaluating the prognosis and guiding the reasonable comprehensive treatment.
