Objective:To explore the plasma apelin-12 levels by ELISA in the patients with essential hypertension(EH) and contrast the difference of apelin-12 in the two groups of EH and control group; And then we want to analyze the relationship between plasma apelin-12 level and carotid artery intima-media-thickness,blood pressure,blood lipids,blood glucose in different blood grades.Methods:68 cases were selected for the subjects of EH group. They were divided into three groups according to their blood pressure:EH grade 1 group(n=23),EH grade 2 group(n =22)and EH grade 3 group(n=23); 22 cases were selected for the subjects of control group. Carotid artery intima-media-thickness was measured by ultrasound.Fasting venous blood was drawn to detect apelin-12,fasting plasma glucose(FPG) and blood lipids including total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C,low-density lipoprotein cholesterol (LDL-C). we determined blood plasma concentration of apelin-12 through ELISA and evaluated blood lipids,FPG and so on. Then we analyzed all the data by statistics software of SPSS13.0, expressed the data with (?)±s and analyzed data of the two groups through independent-sample T Test, Pearson corelations analysis and Multiple regression analysis.Results:1. Comparison of blood plasm apelin-12 in the two groups:It showed that the Plasma apelin-12 level in the 3 grades of essential hypertension group ware significantly lower than that in control group (p<0.01), we analyzed every two groups by one-way anove: It has significance between EH grade 1 and the other grades (p<0.01) and we did not found the difference between EH grade 2 and EH grade 3.According to the results, we concluded the higher of the blood pressure the lower of the apelin-12 levels in the subjects. Pearson correlations analysis showed plasma apelin-12 level was negatively correlated with mean blood pressure.2. Comparison of IMT in the control groups and different grades of EH groups:It showed the significance between control groups and different grades of EH groups; we can found significantly difference of IMT in every two groups of EH (p<0.01); And we concluded The higher of the blood pressure the thicker of IMT.3. Comparison of apelin-12 and IMT in the four groups:It showed significance of apelin-12 and IMT between EH and control groups; Pearson correlations analysis displayed that plasma apelin-12 level was negatively correlated with IMT. 4. Pearson correlations analysis between apelin-12 and other indexes:Plasma apelin-12 level was negatively correlated with SBP,DBP and mean blood pressure in EH group, and negatively correlated with TG,TG and LDL-C in control group;5. Multiple regression analysis of apelin-12 level:in the condition of that the apelin-12 level was to be dependent factor and age, SBP, DBP, PP, mean blood pressure,TC,TC,HDL,LDL were to be indendent factors, It showed that mean blood pressure was independent risk factors to affect the plasma apelin-12 level and none factor affect plasma apelin-12 level in control group.6. Pearson correlations analysis between IMT and other indexes:It showed that IMT was positively correlated with SBP,DBP,PP and mean blood pressure in EH group.7. Multiple regression analysis of IMT:in the condition of that the IMT was to be dependent factor and age, SBP,DBP,PP, mean blood pressure,TC,TC,HDL,LDL,FPG and apelin-12 were to be indendent factors, It showed that SBP,DBP and TC ware independent risk factors to affect the Plasma IMT and none factor affect IMT in control group.Conclusions:1. Comparing with control group, the apelin-12 level was lower than it in EH group; And it showed that apelin-12 level was negatively correlated with BP and IMT;2. It is possible that the decreasing of blood plasma apelin-12 level can be one of mechanism of EH.3. It showed that SBP,TC,IMT and PP were important factors to affect the plasma apelin-12 level in EH patients.4. It showed that SBP,DBP and TC ware independent risk factors to affect the plasma IMT in EH patients.
