Objective:1. To detect the expression retinoic acid receptorαand retinoic acid receptorβin ovarian cancer cell lines SKOV3 and ES2, and to measure the mRNA levels of RARαand retinoic acid receptorβbefore and after using all-trans retinoic acid (ATRA)treated.2. To investigate the effect of ATRA on vitro invasion of SKOV3 and ES2.3. To detect the expression of retinoic acid receptorαand retinoic acid receptorβin normal ovarian tissues and ovarian tumor tissues, and the correlation with several clinicopathologic characteristics.Method:1. Expression of retinoic acid receptors was detected in SKOV3 and ES2 by using immunocytochemistry.2. RT-PCR was used to measure the mRNA levels of RARα,RARβbefore and after using all-trans retinoic acid (ATRA) treated.3. The invasion of cell lines was evaluated using 24-well matrigel invasion chambers.4. Expression of retinoic acid receptors was detected in 20 normal tissues.20 benignin tissues,20 bordline tissues and 52 ovarian cancer patients by using immunohistochemistry.Result:1. RARα,RARβare positive expression in SKOV3, ES2.2. The expression of RARα,RARβwould be increased after treatment of ATRA on ES2 but not SKOV3.3. The invasion of treated ES2 cells was significantly lower than that of untreated ES2 but not SKOV3.4. The positive expression of retinoic acid receptorα/RARβin normal tissues benignin tissues, bordline tissues and ovarian cancer patients were 80.77%/81.48%,50%/45%,15%/30%,20%/30% respectively. The expression of retinoic acid receptorα,RARβwere related to the histological type,FIGO stage, differentiation and so on (P<0.05).Conclusion:The expression of retinoic acid receptorα,RARβwere higher in ovarian cancer.and may be associated with ovarian carcinogenesis. The mRNA levels of RARα,RARβof ovarian cancer cell lines after treatment with ATRA increased and the invasion of the cells decreased indicated that ATRA may inhibit the invasion and metastasis of ovarian cancer by the way of retinoic acid receptors. While SKOV3 cell lines did not have significant change in mRNA levels of RARαRARβand vitro invasion may imply that serous ovarian cancer may resistant to ATRA.
