Objective: To observe the effect of recombine fusion protein IL-3-PE38KDEL on AML cell lines: HL-60(IL-3R positive),NB4(IL-3R negative) , on their generation and apoptosis.Methods: HL-60 and NB4 were grouped respectively during log phase growth: group A(add IL-3-PE38KDEL), group B(add IL-3 and IL-3-PE38KDEL), group C (control group). First, the inhibition ratio and the IC50 were detected by MTT. Then , each A,B group were added an IC50 dose of IL-3-PE38KDEL,calculate the inhibition ratio; To count living cells for 5 days and draw growth curve; Last, the cell cycle and apoptosis were detected by flow cytometry.Consequence: The concentration of IL-3-PE38KDEL was 10μg/150μl, the inhibition ratios were: HL-60-93.4%, NB4-31.6%,after 84 hours. When the concentration was IC50 , the mean inhibition ratios of group A,B were: HL-60(50.2%,32.2%,P﹤0.05),NB4(48.56%,49%,P﹥0.05 ),the mean apoptosis rate of group A,B were HL-60(19.71 %,9.39 %,P﹤0.05),NB4(8.46 %,8.0 %,P﹥0.05,). A majority of cells of HL -60 were arrested in G1/S phase。 Conclusion: IL-3-PE38KDEL can inhibit the generation of the IL-3R positive AML cell lines HL-60,and induce apoptosis. But weak to the IL-3R negative AML cell lines NB4 .
