【Purpose】MicroRNAs (miRNA) have emerged relatively as a new class of small, non-coding RNAs that regulate gene expression and have been identified to be very important in many aspects. Recent studies show that they are very important regulatory molecules in regulating cancer proliferation and metastasis. To predict the regulating miRNA involved in human tumor and deeply study the biological functions which miRNA involved through regulating the expression of target genes.[Experimental Design]The possible interaction between miRNA and proliferation was investigated using MTT and colony forming assay. The impact of processes on invasion by the changes of miRNA was investigated using transwell invasion assay. The target genes were investigated using mRNA expression array combining with bioinformations.And the targets were further conformed by GFP sensor assay. The effect of mRNA being regulated by miRNA was investigated using mRNA expression array and RT-PCR.【Results】Through colony forming assay, we found miR-20a can improve the processes of cell proliferation. The transwell invasion assay showed miR-20a can promote cell invasion and metastasis. We found two candidate target genes by using mRNA expression array combining with bioinformations.And the two target genes were successfully conformed by GFP sensor assay.RT-PCR also confirmed the effect of miR-20a on the two target genes.[Conclusion]Through our studies, we successfully found and identified two target genes of miR-20a, and confirmed the impact of miR-20a on regulating the processes of cell proliferation and invasion.Further study of miRNA may lead to novel concepts in the diagnosis and treatment of human tumor.
