Objective1.To explore the possibility of CGRP given intranasally in improving the behavioral and neurological function of rats after SAH.To investigate the injury of hemolysate to the basic artery and craniotomy microl arteries in rats2.To investigate the protection of the calcitonin gene-related peptide(CGRP) given intranasally in brain injury in rats after subarachnoid hemorrhage(SAH).3.To investigate the possibility of the calcitonin gene-related peptid e(CGRP) given intranasally in relieving the cerebral vasospasm(CVS) in rats after subarachnoid hemorrhage(SAH).4.To investigate the effection of the calcitonin gene-related peptide(CGRP) given intranasally in the system of NO-cGMP in rats after subarachnoid hemorrhage(SAH).Methods1.In this study,adult Wisar rats were used as experimental animals.Cisterna injection twice of freshly autologous arterial blood was used to induce SAH in rats.The rats were divided into four groups:Control;SAH;SAH+IN NS;SAH+IN CGRP.2.Right craniotomy and BA was exposed,drip hemolysate to the surface of BA and micro arteries,Somatotype microscope and livingbody circulation measure system were used to observe the diameter of the artery3.Three days after the second injection,rats were sacrificed.Making brain paraffin sections, detect the diameter of brain microl arteries which is under 200μm and 500μm of cerebral cortex by H-E staining,detect the neuroinjury of hippocampus by Nissal staining.4.Investigate the expression of eNOS mRNA,iNOS mRAN and sGC mRNA in brain arteries by RT-PCR.Investigate the containing of NO in brain arteries by chemical methods.Investigate the containing of cyclic guanosine monophos-phate(cGMP) in brain arteriesResults1.By H-E stain,the loss of neuron,karyopyknosis in partial neurons and anachromasis were fond in SAH group and SAH+IN NS group.Diameters and cross section area of BA were remareablely decreased.The symptom of SAH+IN CGRP group was reliever than SAH group.2.The expression of iNOS in SAH group and SAH+IN NS group were decreased remarkably than the control group,while the expression of iNOS in SAH+IN CGRP group was the same as control group.The expression of eNOS in SAH group and SAH+IN NS group were increased remarkably than the control group,while the expression of iNOS in SAH+IN CGRP group was the same as control group.3.The level of cGMP in SAH group and SAH+IN NS group were decreased remarkably than the control group,while the level of cGMP in SAH+IN CGRP group was higher than control group.4.The level of NO in SAH group and SAH+IN NS group were decreased remarkably than the control group,while the level of NO in SAH+IN CGRP group was higher than control group.Conclusion1.CGRP via intranasal pathway can relieve cerebral vasospasm in rats after subarachnoid hemorrhage.2.CGRP via intranasal pathway can protect ischemic injured of the brain of the brain of rats after SAH leading CVS.
