Influence Of Bupleurum Sanseng Capsule On Myocardial Ca2+ And The Expression Of Serca2a In The Rat Model With Myocardial Ischemia And Cardiac Arrhythmia

Objective:To discuss the influence of Bupleurum Sanseng capsule on myocardial ischemia and arrhythmia in myocardial SERCA2a Ca2+ concentration in rat model and its anti-arrhythmia mechanism.Method:With myocardial ischemia reperfusion rat model,60 wistar rats were randomly divided into six groups, sham operation group, model group, Bupleurum Sanseng capsule with small, medium and large dosages, and Propafenone group. After one week of adaptive feeding, sham operation group and model group were administered the same dose of distilled water by lavage, Bupleurum Sanseng capsule with small, medium and large dosages was administered 1/2 equivalent dose, equivalent dose, and double equivalent dose, and Propafenone group was fed propafenone with dosage 200mg/kg. After 7 days,â…¡lead ECG were recorded, Ca2+ concentration was detected on the fluorescence spectrometer by taking rat myocardial cells load fura2/am fluorescent probes, and expression of the vitality of myocardial SERCA2a mRNA was recorded by using RT-PCR method.Results:After modeling, arrhythmia occurred in sham operation group, in model group arrhythmia occurred significantly earlier, compared with the Bupleurum Sanseng capsule with medium and large dosages, where there was significant statistic difference (P<0.05). Arrhythmia duration of Bupleurum Sanseng capsule with medium and large dosages was shorter than the model group, which had significant statistic difference (P<0.05). But there was no significant statistic difference between Bupleurum Sanseng capsule with small dose group and model group(P>0.05), and no significant statistic difference was shown between Bupleurum Sanseng capsule medium dose group, large dose group and propafenone group (P>0.05). There was significant statistic difference with intracellular free Ca2+ concentration in model group, compared with the sham group (P<0.01), and as well as with the Bupleurum Sanseng capsule medium dose group and large dose group (P<0.01). There was no significant difference between the model group and Bupleurum Sanseng capsule with small dose group (P>0.05). And no significant statistic difference was shown from Bupleurum Sanseng capsule with medium dose group and large dose group to propafenone group (P>0.05). SERCA2a mRNA expression of model group significantly decreased compared with sham operation group (P<0.01). As to Bupleurum Sanseng capsule with small dose group and model group, no significant statistic difference was shown (P> 0.05). Bupleurum Sanseng capsule medium dose group and large dose group could enhance the expression of SERCA2a mRNA, compared with the model group and Bupleurum Sanseng capsule with small dose group, which had significant statistic difference (P<0.01, P<0.05). But there was no obvious statistic difference between Bupleurum Sanseng capsule with medium dose group and large dose group and propafenone group (P> 0.05).Conclusion:Bupleurum Sanseng capsule can enhance the expression of SERCA2a, reduce intracellular Ca2+ concentration, and then reduce the onset of arrhythmia, which plays a role of anti-arrhythmia.
