Two-photon excited fluorescence (TPEF) and second-harmonic generation (SHG), consisted of the nonlinear optical microscopic imaging technique, it combines with the advantage of confocal microscopy. The images and spectra of the biological tissue can be simultaneously got in backward detect. This nonlinear optical microscopic imaging technique can provide more accurate medical basis about the early clinical diagnosis.And Optical coherence tomography (OCT) is a noninvasive morphological method. It allows high-resolution imaging in vivo. An OCT can be used to measure the optical parameters and the structure of tissue.In this paper this two technique were applied in the actue Myocardial ischemia.Firstly,the progress of Myocardial ischemia was reviewed,the structures of cardiac muscle tissue was introduce by anatomy.The cellular level structures was analyzed for better understanding of Myocardial ischemia.Secondly,introduce the OCT technique and two-photon fluorescence technique Last, Due to the important application of left anterior descending corronary artery in pathology research of rat cute anoxemia of cardiac muscle,a cute anoxemia model of rat is established using ligation of the left anterior descending branch in this study.the front wall image of left ventricle and the changes of penetrate depth and Scattering coefficient as time in vivo are attained by OCT, and the front wall Fluorescence image and spectral imaging of left ventricle in different time are attained by TPEF. It can be used to analyze intrinsic species of cardiac muscle, and it can also be used to investigate metabolism of cardiac muscle cells.The experiment results show that scattering coefficient obtained from OCT images and two-photon excited fluorescence spectrum images show a high degree of relevance with histological study and serological test, that means those images have highly relevant with the extent of myocardial ischemia. All of those evidences prove that OCT technique and two-photon fluorescence technique may prove useful to clinical diagnostics as well as to basic biological research.
