ObjectiveTo observe the effect of cholecystokinin-octapeptide(cck-8) repairing the sciatic nerve defects in nerve regeneration indoor, study the ipossible mechanism of repairing rat sciatic nerve defects by cholecystokinin-octapeptide (CCK-8) artificial nerve,providing the basis for clinic using..Methods48 Wistar rats were divided into three groups randomly:Test group included group A (CCK-8 was injected into the PLGA tube) and group B(CCK-8 was injected into the abdomen), control group was group C(physiological saline was injected into PLGA tube).with 16 in each.One sciatic nerve was transected and sutured epineurium with PLGA tube immediately,which was made into nerve regeneration chamber.The rats of group A were treated by CCK-8(56nmol/k) into PLGA tube one time, the rats of group B were injected with CCK-8(8nmol/k) one time per day,7 days total.And control groups received physiological saline of the same volume one time.At the 8th and 12th week, Comprehensive restoration of the sciatic nerve using seven indicators of level of assessment: exercise behave and morphologic changes of regenerated nerve were observed and the recovery rate of the sciatic function index (SFI%) was measured by using the methed of gait. The recovery rate of the complex muscular action potential(CMAP%) and the motor nerve conduction velocities (NCV%) were measured with Nerve-Electrophysiological instrument. The histological structure ofregenerated nerve were observed under electron microscope.And the regenerated nerves were sectioned histologically and stained with hematoxylin and eosin,also firmly green myelinic membrane dyeing to observe.Then, measeure the recovery rate of myelinated nerve fiber counts Measure the recovery rate of wet weight gastrocnemius muscle. Results All the 48 enrolled rats was involved in the result analysis..(1) Exercise behave:At eighth week the hind limb muscle of rats of three group were atrophy, joint stiffness and toe could not reflexed,At 12th week the moving and the hind muscle of group A were best, toe could reflexed.and group B is better than group C. General morphology of regenerated nerve:At 8th week the PLGA tube adhesions with the surrounding tissue, the regenerated nerve grew through the tubes of group A and B, the nerve surface were rough and uneven, group B is obvious,but the regenerational nerve did not pass through the tube of group C. At 12th week the sciatic nerve of group A grew from proximate into distal end,surface and thickness were normal basically,nerve anastomosis had no adhesion with surrouding tissues; the regenerational nerve also grew into distal end,but was thicker than that of group A,surface was rough and uneven, nerve anastomosis had adhesion with surrouding tissues.the regenerational nerve of group C basically grew into distal end, obviouslly thicker than the group A,surface was obviously rough and uneven, nerve anastomosis had notabally abdohesion with surrounding tissues.(2)The recovery rate of SFI were that:-51.04±6.77% in group A,-61.12±8.77% in group B and-72.87±5.85% in group C at 8th week;-34.15±5.57% in group A,-42.29±6.72% in group B and-52.99±7.80% in group C at 12th week.The difference between three groups were significant statistically.(3) Result of Nerve-electrophysiology:at 8th week NCV% and CMAP% of group A were 42.62±0.43% and 64.16±0.98%,that of group B were 35.73±0.28% and 55.28±0.89%,and group C were 23.53±0.76% and 34.97±0.71%,there were difference statistically between three groups.At 12th week, NCV% and CMAP% of group A were 57.23±0.43%and 92.60±2.63%,that of group B were36.26±0.29% and83.40±3.00%,and group C were 23.50±0.61% and 64.01±1.28%.there were difference statistically between three groups.(4)The observation of light microscope and electronmicroscope:at 4th week regenerated fibers in group A were denser and more regular than that in group B, and that in group B were denser and more regular than that in group C.At 12th weeks,the fibers in group A were more regular and homogeneous and myelin sheaths were thicker than that in group B,and that in group B were more regular and homogeneous and myelin sheaths were thicker than that in group C. (6).Quantitative analysis of the recovery rate of myelinated fiber population:at 8th week in group A:63.24±6.65%,in group B:52.08±3.37%;in group C:37.69±6.37%;at 12th week, in group A:81.59±3.24%,in goup B:72.48±5.37 %,in group C:62.37±7.91%.The difference between three goups were all significant statistically.(7)The recovery rate of wet weight of gastrocnemius muscle:at 8th week,in groupA:65.45±1.20%,in group B:60.50±1.44%,in group C:53.05±0.74%;at 12th week,in groupA:78.14±0.38%, in goup B:66.32±1.70%,in group C:54.62±1.04%.The difference between three groups were all significant statistically.Conclusions1 The administration of Cholecystokinin-octapeptide in nerve defects can effectively promotes regeneration of peripheral nerves,more help to improve the limb functions, the role is better than that injected in abdomen.2 This study provides an experimental evidence for clinical administrating Cholecystok-octapeptide to repair the peripheral nerve defects,using it as one kind of artifical nerve material...
