ObjectiveTo investigate the effects of Promote Nerve Regeneration Factors Compound(N6) on expression of malondialdehyde(MDA) and Caspase-3 in experimental rats of whose spinal cord were transect completely, and to approach preliminarily the effects of which on protecting the injuried spinal cord from secondary injury.Methods48 wistar rats were randomly divided into four groups:control group, experimental group, methylprednisolone group and sham-operated group.15 min after transecting T10 spinal cord, Normal saline(NS) and N6 compound were given to the injuried site of 166μl/kg respectively in control group and experimental group,while methylprednisolone(MP) was injected intraperitone-ally of 30 mg/kg in methylprednisolone group, and rats were untreated in sham-operated group.6 rats of each group were sacrificed and the tissue samples were obtained at 24 hours after injury. MDA,a produce of lipid peroxidation (LPO),in broken ends of cord and blood serum were estimated by thiobarbituric acid(TBA) test.Another 6 rats of each group were perfused at 72 hours postinjury,and the expression of Caspase-3 in broken ends were detected by immunohistochemical study while the pathologic changes were detected by HE method.ResultsIn experimental group and methylprednisolone group,MDA of broken ends and blood serum were significantly lower than the control group at 24 hour after injury,the difference was significant (P<0.01). and the quantities of Caspase-3 immunopositive cell located in anterior horns of gray matter were significantly reduced compared with the control group at 72 hour postinjury, the difference was also significant (P<0.05). moreover the difference between the experimental group and the methylprednisolone group was significant(P<0.05).ConclusionN6 compound could protect the spinal cord from secondary injury caused by LPO and apoptosis,and the effects of which were better than NS and MP.
