In order to develop safely and effectively pre-H9 subtypes of avian influenza vaccine,we studied the aluminum hydroxide adjuvant vaccine of H9 subtype Avian Influenza from the safety, effectiveness, duration, shelf life.First of all, explored the antigen concentration in multiples and ingredient in this experiment. 3-fold concentrated antigen and the antigen content at 60% which made the effectiveness of the vaccine was the best were determined. In accordance with this ingredient, prepared a batche of the aluminum hydroxide adjuvant vaccine of H9 subtype Avian Influenza( S01). Then the vaccine conducted the following test. 1. The results of physical properties test and sterile test were qualified. 2.To SPF and broiler chickens, made a 0.5ml, 1ml, 2ml of different doses of vaccine safety testing, test results show that the vaccine for SPF and broiler chickens are safe. 3. The smallest dose immunization experiments show that 0.3 to 0.5ml dose immunization could induce SPF chickens a high level of HI antibodies and chicken reactive immune protection rate could be drug up to 100%; show that 0.4 and 0.5ml dose immunization could induce broilers chickens more than 5log2 of the antibody and the reactive drug protection rate could be up to 100%. 4. to the effectiveness test of the vaccine, results showed that the vaccine in SPF chickens on HI antibody levels generated by higher than broilers. 5.For the duration of the vaccine was tested, results showed that the vaccine effectiveness in SPF chickens can be maintained on 4m and however the vaccine effectiveness could not sustain in the broiler on 4m.. 6. Shelf life of the vaccine was tested, showing vaccine at 2~8℃preservation of 12m.In short, the aluminum hydroxide adjuvant vaccine of H9 subtype Avian Influenza were safe , but the effectiveness of the vaccine in broilers were less than in SPF chickens. The question is to be explored further.
