| Apomictic monosomic addition line M14 was obtained from the hybridization and intercross of Beta vulgaris L. and Beta corolliflora Zoss. M14 with the No.9 chromosome of Beta corolliflora Zoss. (VV+1C,2n=19) was found having a chromosome transmission frequency of 96.5%. It was proved that M14 served as ideal materials for cloning apomixes genes. Studying the specifically expressed genes in M14 could make the research of the excellent properties in M14 a higher value and significance.Both BvM14-MADS box gene and BvM14-Rab gene were specifically expressed genes in M14. The full-length cDNA fragments of these two genes were transformed into the model plant tobaccos respectively in our laboratory. In order to study the hereditary character further of the transgenic tobacco plants, two trangenic tobacco plants and their progenies which introduced BvM14-MADS box gene and BvM14-Rab gene were used as materials to perform molecular detection, their phenotypic observation and biostatistics analysis.Foreign gene could be inheritance to the next generation. However, it varied in lines and generations. The transgenes were transmitted to progeny according to Mendelian and in some cases non-Mendelian patten.84.6% NPT-â…¡gene and 82.8% BvM14-MADS box gene in BvM14-MADS box transgenic progenies were accordance with Mendelian segregation. Whereas,15.4% NPT-â…¡gene and 17.2% BvM14-MADS box gene were according to non-Mendelian segregation.84.0% NPT-â…¡gene and 74.1% BvM14-Rab gene in BvM14-Rab transgenic progenies were accordance with Mendelian segregation.16.0% NPT-â…¡gene and 25.9% BvM14-Rab gene were according to non-Mendelian segregation.Linkage analysis of the selection gene and target gene showed that these two genes cooperated fine with each other in the genome of a single plant, while segregating separately in several plants.The morphology analysis of transformed plants showed that BvM14-Rab transgenic progenies emerged a series of abnormal phenotypes, such as geotropism, a lower height, more shoot branching, a decreasing inflorescence and a lower fertility. BvM14-MADS box transgenic progenies emerged shoaling of petal color, longer stamen and pistil, delaying of florescence and a lower fertility.It was proved that the foreign genes could inherit steadily in transgenic tobacco plants by molecular detection, analysis of the phenotype changes. And the homologous transgenic plants were obtained in T3 generation. |
