In the stuty, we use the sheep as the reaser ch object, on the basis of observing the sheep ovary using histological techniques, compare the two effects of getting ovarian surface follicles oocytes in the extraction and cutting methods, and do maturation study in vitro, observe the genera] morphology and lipid content in different stages of the development of sheep oocytes.The experimental results:Sheep ovaries are generally oval-shaped, the number of primordial follicles and primary follicles are fewer than other livestock, three follicular phase can see clearly the formation of cumulus.Compared with the suction wears, the number of available oocytes collected by cutting method and the maturation rate and diffusion rate of the oocytes after culture were significantly higher than the aspiration (P<0.05), And denuded oocytes of cutting method was lower than aspiration.During the maturation process of sheep oocytes in vitro, adding 0.095mg/mL cysteine can significantly increase the maturation rate of oocyte in vitro.Sheep oocyte lipid content is less, Oh development of oocytes, the density of lipid droplets was Uniform, dense,8h lipid droplets slightly reduced, lipid droplets became larger,24h,30h Lipid droplet sizes and uneven density in blastomere. |
