In this study five or six generations F1,F2,backcrosses and their parents P1 and P2 were used to study the inherence of flower color and flower color pattern of pansy and hybrids of viola and pansy;We had established primary system of rapid propagation and regeneration of pansy to provide the technical platform for creating male-sterile lines using the genetic engineering.The results were presented as following:1.The results of color on the petals of the tristimulus values measured by colorimeter shown that petal color of 11 inbred lines of Pansy and an inbred line of viola can be roughly divided into the following five colors:yellow,purple,purple blue,red and white.The colors of flower color pattern of Pansy had narrow distribution,mainly distributed in the purple and were darker than patals'.2.Most of petal color of F1 hybrids showed a continuous variation between two parents'.Others had a flower coloring similar to one of the parents or an intermediate color between the parental flower colors.F2 generation segregated deviating colors except that shared parents with same color.The hybrids of pansy and viola shared the similar results with hybrids of inbreed lines of pansy.3.Five or six generations F1,F2,backcrosses and their parents P1 and P2 were used to study the inherence of flower color pattern of pansy and hybrids of viola and pansy.It was found that blotch was completely dominant to the stripe pattern,and that stripe pattern was completely dominant to pure color.Blotch was controlled by two genes in the presence of stripes.4.In this study inbred line 22-8 was used to set up the rapid propagation system,it was found that MS+2.0 mg/L BA+0.1 mg/L NAA was found to be the best initial medium,that MS+2.0 mg/L BA+0.1 mg/L NAA was the best Subculture medium,and that 1/2MS+0.1 mg/L IBA was the best medium for rooting.Phenomenon of vitrification was usually seen in the Proliferation process,and the result of rooting was not satisfactory.5.In this study inbred line 22-8 was used to set up the direct regeneration system. There were not plantlets regenerated from in vitro-grown leaf,petiole and intermodal segments explants on MS basal medium supplemented with different concentrations of BA and NAA,ZT and NAA.Plantlets were regenerated from in vitro-grown explants of genotypes of pansy on MS basal medium supplemented with different concentrations of TDZ and NAA from petiole and internodal segments explants.The highest regeneration rate(56.7%) were obtained from internodal segments explants cultured on MS basal medium supplemented with 1.0 mg/L TDZ and 0.1 mg/L NAA.The highest regeneration rate(25.0%) and the largest number of shoot clumps per regenerating leaf(2.7 shoot clumps/explant) were obtained from intemodal segments explants cultured on MS basal medium supplemented with 0.5 mg/L TDZ and 0.1 mg/L NAA.AgNO3 inhibited regeneration from internodal segments and petiole explants of pansy.Plantlets were regenerated from in vitro-grown explants of another genotype 4×15 on MS basal medium supplemented with TDZ and NAA.
