Sea buckthorn, riching effective components including vitamins, flavonoids, polyphenolics, organic acids, etc, have so multiple biological and pharmacological activities including antioxidation, protective effect on cardiovascular system, improve immunity, anticancer, and so on. It is important to have edible value, medicine value and ecology value. People are only interested in buckthorn flavonoids and sea buckthorn oil research and do not put emphasis reaearch on the other very important effective components—proanthocyanidins. Proanthocyanidins, are polyhydroxy phenolic compounds, have more biological and pharmacological activities and profound potential futureBased on the former work, the paper optimizes the extraction technology of proanthocyanidins from sea buckthorn, considered the effect on extraction rate of proanthocyanidins from sea buckthorn combining degreasing time, alcohol consistency, solid-liquid ratio, pH value, extraction temperature, extraction time, designed experimental scheme using U11(1110) uniform designs table and obtained regression equations: y=0.013303618×a2-0.411850513×a-0.001343630×b2+0.156436878×b-0.015037670×c2+0.246618524×c-0.113659826×d2+0.121947603×d-0.002176696×e2+0.041272882×e-0.000086656×f2-0.008098396×f-0.002862893×d×e+0.000013987×a×b×e+0.000067755×b×d×e+0.000149079×c×d×e-0.000006776×d×e×f+12.235182420. Base on this, predicted the extraction rate is the maximum that is 17.7063 percent which improve of 5.2828 percent than before. In addition, obtained the proanthocyandins production of oligomer and polymeride by extraction separation using acetic ether. D3520 macroporous adsorption resin and 30% ethanol solution eluting are more ideal purification method of proanthocyanidins from sea buckthorn, and the purity of the maximum is 88 percent. This paper verificated and compared the stability of proanthocyanidins from sea buckthorn seed and grape seed on the environment including pH value, temperature, oxidant and reductant, confirmed free radical scavenging activity of proanthocyanidins from sea buckthorn.In addition, the paper screened out and verified Penicillium which can degrade proanthocyanidins from sea buckthorn seed. Penicillium screened out having capacity of degradting polymeric proanthocyanidins were determined.
