The nitric oxide (NO), one kind of inorganic minor numerator, is commonly found in animal cells, plant cells and bacteria as well. NO has long been known as an notorious environment pollutant, which contaminates air, develops rain formingacid, and destroys the ozonosphere. Until recently, People were ignorant of the fact that this kind of molecule plays an important role in the body of living things. Recent scientific researches indicate that NO is one kind of living things activity molecule, a signal molecule commonly existing anhistozoa, bacterium, yeast, animal and plant. Many researches have been conducted on the function of NO in the mammal and plant, but the researches concerning plain core microorganism producing NO have hardly been reported yet. The current experiment has screened NO bacterium in soil through various methods, and carried out measurement in such respects as growth and physiology characteristic. At the same time it has also studied the NO production mechanism of NO bacterium and the possible influence on the bacterium living things amounts in soil environment. The major research results are as follows:1. With different medium and train way, over 400 bacterial strains are obtained from soil and sludge. 66 NO producing bacteria are obtained through screening structuring gfp, the NO bacterial strain cooperated by the Laboratory of USA Cornell University and our laboratory. We get 10 NO antinitrifying bacterium after getting rid of the nitrification bacterium ultimately.2. The significant effect that temperature and pH have on the NO bacterium's abilities to produce NO. In logarithm growth period of Bacterium NO, the effect of the temperature and pH is the most obvious. B30's growth is the fastest and the NO production also reaches the maximum at 30℃and pH7.5. It indicates that the fastest growth period of B30 is also the maximum NO production period.3. The analysis of B30 NO production mechanism through using 4 kinds of nitric oxides enzyme (NOS) depressor experiments. The result indicates that L-NMMA (general NOS restraint )and Spermidine (nNOS depressor) have obvious restraining effect on Chryseobacterium. It has also suggested the NO production mechanism of NO bacterium and that NO bacterium takes L- Arg as front body matter, which produces NO under nitric oxide enzyme.4. The effect of NO on soil environment middle bacterium number by inoculating with NO bacterium to soil. The result indicates: Inoculation with NO producing bacterium obviously reduces the culture of bacteria quantity in soil, and NO producing bacterium may have adjustment function for soil bacterium community structure.
