The roots of Glycyrrhiza uralensis Fisch, commonly known as liquorice, have therapeutic properties. The main component is triterpenoid saponin thetriterpene glycyrrhizinic acid(GL) which is between 3% and 8%.Another bioactive component is glycyrrhetinic acid (GA) which is mainly used in pharmaceutical and cosmetic fields as anti-inflammatory, anti-ulcer activities and anti reddeningagents. Furthermore, the anti-hepatotoxic and the hypolipidemic activity of GA have been demonstrated. But the content of GA in liquorice is only about 0.1%.The main contents and results include:Ⅰ screening, identifying of strains which can transform GL with high ability. Based on the entironment of strains, GL was used as the screen plate to separate and purify the strains with high hydrolyzation.The method was simpleness and effective. We have easily isolated HC—12 from soil samples of wild Glycyrrhiza in Xinjiang province. Through the analysis of TLC and HPLC Results, HC—12 is Aspergillus oryzae with high ability on conversing of GL.Ⅱ optimizing of the fermentation conditions of transformation with HC—12. According to the result of single fact and orthogonal test, the concentration of GA reached the highest (2.22 g·L-1) in the following condition: 100 mL of culture medium in a 250 mL flask by adding 7.5g substrate at hour120, and incubating at 32℃ with agitation speed at 180 r·min-1. we also have drawn the dynamics curve of fermentation.Ⅲ isolating, purifying Glycyrrhetinic acid from the liquid fermentation extracts. We have used static method and the technology optimizing disposition of thesystem factors with all over feedback control to screen out a macroporous resin H103.The best technics is that : consistence of GA1.5mg·mL-1,NaCl 1.5%, adsorption speed 2 BV·h-1, adsorption pH 6.5, resolving pH 8, resolving speed 1 BV·h-1, it can get 46.64mg·g-1.Through the methods of macroporous resin and silica...
