Asparagus officinalis is recognized as a traditional medical plant for broad pharmacological activities. There are some chemical components in Asparagus officinalis which has many function of anti-tumour, reduce blood fat, immunomodulation, fight against senium, anti-fatigue, antioxidant and so.In this paper we isolate flavonoids from Asparagus officinalis by means of 70% ethanol with ultrasonic wave treatment, the content of total flavones in Asparagus was determined by three wavelength spectrophotometry, and standard rutin sample as the internal standard were employed to determine the content of flavones (rutin) in Asparagus by RP-HPLC.Total flavonoids were extracted from Asparagus by 70% ethanol solvents with ultrasonic wave treatment. Based on the reaction of flavonoids with AlCl3 to produce stable complex, we determined the flavoniods by three wavelength spectrophotometry. The extracting efficiencies of 70% ethanol is 0.177%. Moreover, the stability of the flavoniods is very well. The results indicate that the accuracy of this method is more advantageous than tranditional spectrophotometry method.Agilent Eclipse XDB-C18 column, elution buffer of methanol-water-phosphate acid (55:44.5:0.5) solution as mobile phase and standard rutin sample as the internal standard were employed to determine the content of flavones (rutin) in Asparagus by RP-HPLC. The flow rate was 0.7ml/min and the detection wavelength was 390 nm. The results showed that the base lines of all components were separated well. It was found that there is a linear relation between the peak area and concentration of Rutin in range of 0.07 to 0.7μg/m with a linearity correlation coefficient of 0.99941. The regression equation was A=15176C-10.388. A recovery rate of 101.051% with relative standard deviation (RSD) of 3.306% was got in the study . The RSD of time and peak area in precision test was 0.199% and 1.24%, respectively. Our study suggested that this method was simple, accurate, and also suitable for the quantitative analysis of flavones in Asparagus.We choosed spectrophotometer method to study the extract solution and flavonoids ofAsparagus officinalis on reactive oxidative free radical. In vitro, it was shown that the extract solution and flavonoids were found to have significant capacity of scavenging free radical in a concentration of dose-dependant manner. The scavenging effects on hydroxyl free radical and superoxide anion free radical by 5% extract solution and flavonoids with the concentration of 5mg/ml were above to the concentration of 20mg/ml of manna. In addition, a significant inhibition of malondialdehyde(MDA) generation spontaneously in liver homogenate is also oberserved in vitro.All above results will provide some reference for making more exploitation on Asparagus officinalis.
