Songliao Black Pig And Western Henan Fat Tail Sheep Fibroblast Cell Lines Of The Establishment,

As the basic component of biodiversity, domestic animal genetic resources are very important genebank. It is of economic, scientific and in cultural interests to humankind in terms of food and agricultural production for the present and in the future. Nevertheless, Higher lever of industrialization is realized through raising a high-productive breeds(or hybrids) to replace local breeds. Decline and deprivation of genetic diversity in livestock and poultry breeds took place progressively. Songliao black pig and Yuxizhiwei sheep, which are two important loc-al breeds in China, were sampled and fibroblast cell lines of them were establish- hed and characterized respectively in this research:1. The ear marginal tissues of the Songliao black pig and Yuxizhiwei sheep were successfully cultured by primary explanting technique. Fibroblast cells were separated by trypsin and 5 times subcultured. Morphology and analysis of dynamic growth showed that the population doubling time of cells made with the two breeds were 35.5h and 41.5h respectively. A certain extent number of cells for these two breeds were freezed, and viability examination of these cells showed that viable rates were higher than 89%before freezing and after recovery.2. Observations on morphology, analysis of karyotype, isoenzymes for the two established cell lines were undertaken, and expression of GFP in the cells were carried out. The results showed that for Songliao black pig, the frequency of cell chromosome number to be 2n=38 was 92%;for Yuxizhiwei sheep that to be 2n=54 was 89.5%. The banding patterns of the isozymes of the two enzymeshad significant normal;tests for microbial contamination from bacteria, fungi or mycoplasma were negative;transfer efficiency for the plasmid was high of 37%.The two newly established cell lines make the Songliao black pig and Yuxizhiwei sheep breeds, two national important genetic resources preserved in cell level, as well as will provide an ideal experimental material for the genetic studies on these two local breeds.
