The shoot apex of Cherry rootstock Colt(Prunsavium×Pruns pseudocerasus) was used as explant.Transformation by Agrobacterium tumefaciens with antibacterial peptide gene was studied in this paper.In case of the capacity of transformation to tabaco,we put pSMC to A208SE.9 transformed clcones were obtained by pSMC/A208SE. The results of molecular biological assays with PCR confirmed that the antibacterial peptide genes had been integrated into the genome of cherry.Antibacterial-resistant test and inculating with C58 for the transgenic plants showed that transgenes could be expressed. High transformation frequency could be received when Agrobacterium were re-suspended in activation medium YEB-1 (pH5.6) with concentration of OD600  0.4. It was important to pre-culture explants before infection, and 1 day was the best. The shoot apex from 25 day-old tube-plants were benefit for obtaining transgenic plants. 5~7 min for infection time, 3 days for co-cultivation period ,3~7 days delay selection and temperature of co-cultivation with 24~26℃ were the best transformation condition. The highest transformation frequency was 3.01±0.2%.
