Using two series of the transgenic tobacco expressing cytosolic CaM/CaMBP as experimental materials, we studied the impact of CaM activity on SOD and its isoenzymes.The dissertation is divided into two parts:Part Ⅰ We measured the CaM activity of 10 transgenic tobacco lines expressingnuclear CaM and selected three good transgenic tobacco lines(5C, 5L andl3). In this part, the materials for the next experiment were screened and the optimum Tc-inducible time was confirmed.Part Ⅱ Using the two Tc-inducible expressing cytoplasmic CaM/CaMBP transgenic tobacco lines(Hl-2C, H3-19) leaf disks as experimental materials, we measured the activity of superoxide dismutase (SOD) and their isoenzymes. The results showed that SOD and isoenzyme activities of Tc-inducible transgenic tobacco (2C ) were higher than those without Tc-inducible, and the transgenic tobacco(2C) without Tc-inducible was higher than the control (HO2020). On the contrary, SOD and isoenzyme activities of Tc-inducible transgenic tobacco (19) were lower than those without Tc-inducible, and the transgenic tobacco(19) without Tc-inducible was lower than the control (HO2020). These results were confirmed by ployacrylamide gel electrophoresis (PGE). The results above implied that CaM might play an important role in regulating SOD and isoenzyme activitise of transgenic tobacco. |
