Pig Bronchial Bordetella Pcr For Detection Of The Establishment And Application

Bordetella bronchiseptica is one of main pathogenies of atrophic rhinitis in pig. The importance taches of preventing atrophic rhinitis arc detecting Bb fleetly and washing out carriers early. The designed primers based on Bb flagellin gene upstream sequence were used to PCR, Bb can be amplified 237bp.We established a clinical diagnose method of detecting Bb fleetly. Specificity and sensitivity assay showed that this method had no cross reaction with E. coli, D type Put and S. aureus;112. 5pg template DNA can be amplified .In order to investigate applicability of PCR, 48 clinical samples from swines of Yanbian state were detected by PCR. The result showed that 42 Bb were detected, positive rate of PCR was 87. 5%. At the same time , comparing with conventional bacteriscopy and microamount agglutination, positive rate of bacteriscopy and microamount agglutination were respectively 16:7% and 50%. It indicated that PCR is 5. 25 times than bacteriscopy and 1. 75 times than microamount agglutination. PCR was more fleet and sensitive.PCR had some merit of simple, fleetness, specificity and sensitivity, providing diagnosis of AR for technique . It can be used to diagnosis of latent and earlier period , epidemiology investigation.
