Because dendrobium is a kind of aerophyte, and request forentironment comparatively stringent, outgrowth propagate frequency is highly low(<5%), adding to lasting collection, the wildness plant is close to annihilation. This research first reports theRAPD analysis for the hereditary stability of D. huoshanense and allele-specificdiagnostic for authenticating D. huoshanense.1. To research the hereditary stability in one capsule, and the diversity betweendifferent capsules, which are from D. huoshanense, using three optimal decameroligonucleotide random primers which were screened from 20 primers for DNAamplification, random amplified polymorphic DNA (RAPD) were performed for fivecapsules. The coefficient of genetic similarity are very high, varying from 92.59285%to 100%, and the variation were present in the H1, H9, H10 and H14, but the H23 wasa stable group. Among H1, H9, H10 and H23, the genetic distance were smaller thanthat of the H14. The formation of the rapid propagation system of D. huoshanense isfeasible. At the same time, the results indicated that the variations are in existence in it.The H14 appeared to be the highest diversity compared with other capsules.2. To research the hereditary stability of D. huoshanense which were subcultured7-8 times in the same tissue culture system and come from five different developmentstages, the same three primers and RAPD analysis were used. The result showed thegenetic similarity coefficients are very high, varying from 85.4093% to 98.3606%.The variation of protocorm, germination and monoleaf-stage are rather more notablethan that of bileaf-stage and inflorescence. The variation of the same capsule issmaller. To set up the rapid propagation system of D. huoshanense is feasible.
