In presentswe studied on many factors influencing the propagation in vitro of narcissus for the purpose of setting up efficient regeneration system of Narcissus tazetta var.chinensis Rome. The results showed as follows: It was very important to choose and pretreatsuitable explants for the starting tissue of narcissus in order to decrease pollution rate and improve differentiation rate of adventitious bud and small bulb in the process of culturing narcissus in vitro. The explants which were treated with low temperature could maintain their vitality and original color after incubating, Even the cold-treatment lasted 4 months, the vitality rate of the explants kept in 95%. Moreover, they tended to be induced adventitious bud and differentiation of small bulb. The pollution rate decreased from 95% to 10% after cutting bulbs which have been removed membrane scale leaf and old roots and washing them with flowing water. By observing the tissue culture of explant incubating in different season, we found that the induction rate of small bulb of Narcissus explants incubating from February in 2003 to May in 2004 reached 100%. The small bulbs induced were full and the shapes of them were regular. The explants incubating in other seasons were not suitable. By comparison different incision methods, we found it' was better taking stem plate carrying bulb leaves as explants than any other treatments.The experiments showed that the kinds and concentrations of ex-hormones were key factors that affect tissue culture and regeneration of Narcissus in vitro. The medium containing 0.4 mg · L"1 IBA was better than that of NAA and 2,4-D separately on inducing small bulb as regard to induction quantity and velocity. And the small bulbs induced in this medium were big and regular. But medium with the combination of NAA 0.1 mg · L-1 and 2, 4-D 0.3 mg · L-1 was well suitable for inducing stem and bulblets. If the concentration of hormones were the same,and the concentration of BA reached the 15 mg · L-1, the emergence of bulblets developed well. When the concentration of BA is 10 mg · L-1 ,the callus developed well .But when the concentration of BA reached 20 mg · L-1 ,the quality of callus would become worse and the quantity became less. The effects of the concentration of 6-BA on multiplication of adventitious bud were significant if the leaves were taken as explants. When the BA concentration is 5 mg ·L-1, the multiplication coefficient was 5.1. When the BA concentration in the medium was more than 5 mg · L-1, the multiplication coefficient became decreasing with the increasing of BA concentration.As regard to inducing root of tube seedling of Narcissus, we found that the medium with 0.5 mg · L-1 NAA was best suitable for rooting, and in which the rooting rate is 95.3%,next...
