Application of Genetic Markers in Interspecific Hybrid of Gossypium(Cotton) ABSTRACT Gossypium bickii had some properties such as the high resistence to deseases and insects, glandless seed and glanded plant. In order to utilize the useful traits of G. bickii, the amphidiploid was got from G. arboreum X G. bickii, then some trispecefic hybrids were derived from G. hirsutum X (G.arboreum X G.bickii), (G.arboreum X G.bickii) X G.barbadense. This study was carried out to investigate the morphology, isozyme and molecular marker of the trispecific hybrids. The main results were as follows: The results of morphology showed: he hybrid of G.hirsutum X (G. arboreum X G. bickii) had the same characters as G. hirsutum, G. arboreum, G. bickii. 25% of characters of the hybrid came from G. hirsutum and G. arboreum, 25% from G. bickii, 4.2% from G.arboreum, 12.5% was superior to its parents. ï¹–he hybrids of (G.arboreum X G. bickii) X G. barbadense had the same characters as G. arboreum, G.bickii and G.barbadense. 33.3% of characters of the hybrid came from G.arboreuin and G.barbadense, 12.5% from G.bicki, 8.3% from G.barbadense, 12.5% was superior to its parents. Analysis on peroxidase isozyme of the two hybrids indicated: @ G. hirsutum X (G. arboreum X G. bickii) had 9 bands, and displayed hybrid band parttern. 33.3% of its bands originated from G.hirsutum, G.arboreurn and G.bickii, 11.1% from G.hirsutum, 11.1% of the bands were hybrid bands. ?(G. arboreum X G. biclcii) X G. barbadense had 11 bands, and displayed partially complementary band pattern. 36.4% of its bands originated from G.arboreum X G.bickii and G.barbadense, 9.O%from G.bickii, G.barbadense, 27.3% from G.barbadense. RAPD analysis of the two hybrids using 24 arbitrary primers revealed: total of 131 DNA bands were amplified in the hybrid of G. hirsutum X (G. arboreum X G. bickii). Of them, 65.6% came from the amphidiploid of G.arboreumXG.bicki. total of 145 DNA bands were amplified in the hybrid of (U arboreum X G. bickii)X G. barbadense. Of them, 61.4% came from the amphidiploid of G. arboreum X G. bickii. All of the results of morphology, isozyme and molecular marker showed that genetic substance of the amphidiploid had been transferred into G. hirsutum and G. barbadense, and the hybrids of G. hirsutum X (C. arboreum X G. bickii), (G. arboreum X G. bickii) X G. barbadense were truly trispecific hybrids.
