In greenhouse the pathotype of 41 strains of Vertwilltum dahliae of cotton were studied by maculating the seedlings of 4 different cotton cultivars, EjingNo. 1(S)~ Zhong-12(T) WcnS(R) and TangmianNo.2(R). 41 stains were clustered into 2 groups: defoliating types and nondefoliating types. Amplified fragment length polymorphism (AFLP) was performed on 41 strains of V dahiiae of cotton in search of DNA probes specific for distinguishing between nondefoliating strains and defoliating strains. Specific bands of 433 base pairs(bp) and 1 lObp amplified only from nondefoliating strains were obtained using two pairs , E64 (GACTGCGTACCAATT CGAC) M53 (GATGAGTCCTGAGTAACCG) and E49 (GACTGCGTACCAATTCCAG) M65 (GATGAGTCCTGAGTAAGAG) ,which named EM433 and EM110, respectively. The specific band EM433 was recovered., cloned and sequenced, and 2 primers of Sequence characterized amplified regions(SCAR) markers, 5 - -CGAGACGTCCGAGTC GATATAGC-3 - and 5 - -GTCGTCCTTCTAGTGGGTATCCTCC-3 -, were deviced and systhesised in terms of the result of sequence. 41 strains of V dahliae were amplified specifically, 9 nondefoliating strains and 2 defoliating strains could yield EM.~33, and accuracy percentage rate identified come to upwards of 90%. Accordingly, specific band EM433 could be used for identifying nondefoliating strains. System cluster analysis based on 169 polymorphic bands from results amplified with 8 effective primers to generate adendrogram.4 1 strains of V. dahliae were clustered into 2 groups: 10 nondefoliating strains and 1 intermediate strain; 30 defoliating strams,including 4 control defoliating strains and 26 defoliating strains from northern cotton area. According to dendrogram, We suggest that certain relevant relationship exist in strains of different geographical source.
