Human tissue plasminogen activator(t-PA) is a special effective medicine for the cure of cardiovasculor diseases caused by thrombus.In order to get the transgenic animals which can express t-PA in mammary gland, a series of pBLG- t-PA-bGHpolyA gene clone techniques were used to construct the mammary-special expression vector of t-PA, pBLG- t-PA-bGHpolyA, which consists of 1.0kb bovine ~ -loctoglobulin (BLG) gene 5?flanking region, 2.1kb human i-PA eDNA gene sequence and bGH polyA gene sequence. Hi-PA transgenic mice were constructed by microinjection of a Sail linear fragment of pBLG-t-PA-bGHpolyA(5.4kb) into fertilized eggs0 One thousand and fifty-three microinjected eggs out of one thousand and three hundred and sixty-five fertilized eggs were transplanted into fifty pseudoprenant mice.Twenty-four of these pseudopregnant recipients became pregnant and seventy-nine offspings were produced .PCR and Southern blotting were used to analyse the genomic DNA samples extracted from offspring tails.The result of PCR showed thet nineteen offsprings were positive The result of Southern blotting showed thet four of them were are positive.
