Primary Hemocytes culture of Macrobrachium nipponensis were reported first. Five media and two different osmolalities were compared for their effect on growth of culture cell and the activity of Alkaline phosphatase in intracell. The result showed that M199 (GIBCO) medium was the best, M199 (SERVA) was better, and 472mmo1/Kg was better than 75Ommo/Kg for hemocytes culture of Macrobrachium nipponensis. Using the method developed, studies were carried out to the first subculture from hemocytes of Macrobrachium nipponensis. The optimum conditions for shrimp cells in M199 were determined to be an osmolality of approximately 472mmo1/Kg, 20% fetal bovine serum (FBS), 100U/ml penicillin, 100p~g/ml strptomycin, pH range of 6.9-7.1. Supplements tested included: glucose, vitamine C and metal ions (Cu>, Zn>). The result showed that with supplements of 0.lmg/L Vc and lg/L glucose in basic medium, the cells attached to the surface and grew well. It can be found out that when the concentration of 2?was 2jigIL, the activities of AKP was the highest, and that concentration, the total phosthotus had the highest numerical value. When the concentration of Zn> was 80~tg/L, not only the total phosphorus was the highest, but also the cells grew best. We carried out primary cell cultures from muscle and hemocytes of Penaeus japonicus and electron microscopical observation of hemocytes cultured. When we added 20% FBS, some amino acids and inorganic salts, hemocytes cultured formed a monolayer in a week, but unattached after subcultured. The observation of electron microscopice showed that attached cells became deformed and had some blank blisters.
