For mapping pea comb gene, using 2 pea-combed ((?), from Turfan, China) and 6 rose-combed ((?), from Shihezi, China) chicks as parents, which comb genetypes were proved to be all homozygous by testcross, produced F1 chicks ((?)8,(?)8), which were all walnut-combed, and a group of F2 chicks (494) according to classical genetic principle of Mendal, of which, walnut-combed 295, pea-combed 85, rose-combed 82 and single-combed chicks 32. Statistical analysis by X2 test indicated that the phenotypic ratio of walnut-combed, rose-combed, pea-combed and single-combed chicks was 9:3:3:1, according with classical Genetics theory.Selected 20 chicks with complete family trees and traits record from 494 F2 serum samples, then chosed 13 pairs of microsatellite markers on chromosome 1, which were MCW0436, MCW0019, MCW0428, ADL0319, ADL0150, LEI0174, MCW0112, MCW0018, GCT0049, MCW0058, LEI0071, MCW0007, ALEV1,to locate comb gene and make linkage analysis.The genetic linkage map of chromosome 1 constructed by MapMaker/EXP 3.0 and MapDraw 2.1 software. The results as follows show that:1. Except MCW0112,MCW0007, experimental use of 11 microsatellite markers in the test group showed a higher heterozygosity and genetic polymorphism information content, The population heterozygosity(H) and polymorphism information content (PIC) of groups were 0.3827-0.8533 and 0.6067-0.8356 respectively. Of which, MCW0058 was highest.And all of 11 microsatellite markers (PIC>0.5) were polymorphic.2. Based on the results from research and analysis reported, the preliminary view that the existence pea comb gene on the 1 st of chromosome. Screening 4 microsatellite markers in the chromosome 1 based on known map. It is these markers that were located near short arm of the chromosome 1 and used to do genetic mapping.3. Use reference family in the domesticto map pea comb gene firstly. Therefore, the construction of resource family, and the mapping of genetic linkage map of the comb traits preliminary analysis. Use of maximum likelihood method, the LOD value of MCW0428, ADL0319, LEI0174 and MC W0058 were more than 3, of which, LEI0174 was linked with pea comb gene and the distance between them is 0.12 cM. It laid a foundation for the more precise mapping.
