Study Of Inoculated Moderately Halobacterium Salinarium Salination Cotton Field's Microorganisms Population Constructions

In the present experiment a strain Moderately Halobacteriu-m salinarium,Bacilus.sp.Xjl-05 which was isolated from Xinjiang Aibi Lake, was used as the test strain. Due to study its optimal cultivation condition, single factor and orthogonal experiment had been combinated to research through shake flask cultivate. Bacillus sp.XJ1-05 was used to inoculted to gently salination cotton fileds based on the above results,and investigating the variation of microbial population of bacteria,actinomycetes and fungus in different of vertical soil layer during different cotton growing period.PCR-SSCP molceule marker technology was applied to study soil un-cultivatable mircobial populations at Shihezi Forest Management Station, Xin-jiang. The experiments shows following results:1. Bacillus sp.XJl-05 strain growing into stationary pHase 36h after inoculated which sized.The results also shown that the optimal medium by add 12g/L of yeast powder and 5g/L of ammonium sulfate as well as 7 percentage of sodium chloride, and shake flask cultivate optimum temperature is 47℃,pH9.0 and 220rpm/min.2. After inoculate Bacillus sp.XJl-05,the bacteria and fungus quantility were rapidly increased in a short period.But the quantility of actinomycetes increased slowly after a short time of inoculation.In the three iquid bacterium,the dilute iquid bacterium is more barrer than others.The results suggesting the soil alkalinity is relieved.3. Through SSCP analysis, Bacillus sp.XJl-05 can increase the diversity of bacterias in the soil layer of 5-25cm,but it's no boversily effect for the soil layer of 25-35cm.After inoculate the Moderately Halobacteriu-m salinarium,the major bacterium groups have tiny diffence,and the bacterium community is stabilize.The cluster and diversity index analysis show inoculating...
