| China is the most important agriculture country in the world. Its productivity of agriculture was not only pay seriously role on the people life, but also to the security of world grain reserve. Striped hamster (Cricetulus barabensis) is one of the pest rodents in the north of China. Because of its damagment, the grain loss was about several billion's grain every year. In recent years, the destruction to the grain's productivity and the Steppe and forest ecosystem of Cricetulus barabensis in the drought and semi-arid areaes has upgraded. Athough the environment changes from bad to worse, the research on Cricetulus barabensis is slim. So the research on the C. barabensis reproductive mechanism has significant theoretical role on control their damage.Luteotropic hormone receptor, as one receptor of the gonadotropin hormone, play an important role in mammalian reproduction. In female mammalian animals, luteotropic hormone receptor connect with luteotropic hormone to regulate ovulation, nidation and pregnancy by the system of the hypothalamus-adenohypophysis-ovarian axis; In male, it can promote the interstitial cells secrete testosterone and Sertoli cells differentiation. In order to obtain the gene sequence of luteinizing hormone receptor (LHR) of C. barabensis), the primer was designed according to the homology sequence of golden hamsters (Mesocricetus auratus), mice (Mus musculus) and rats (Rattus norvegicus), a 154 bp LHR sequence had been obtained by PCR reaction. This fragment sequence localized at the 11th exon of 5'and encoded 51 amino acid including partial hinge region and the first transmembrane helical segment (TM1). The hydrophobic value indicates typically a-helix in TM1 and the proportion of Val, Ile, Leu were 9.80%,9.80%,19.61% respectively. Alignment of LHR fragment sequence with that of other species, the nucleotide and amino acid homology were 83%-98%,82%-100% respectively. Although amino acid homology of TM1 was conserved and up to 82%, there is significant distinction on sites 26,41,44,48 between rodents and other species.Testis of C. were collected for paraffin sections; Adjacent serial sections were selected for hematoxylin-eosin staining to confirm that C. barabensis was in estrous. LHR expression quantity in seven tissues was compared by FQ-PCR. The result showed that LHR was all expressed in seven tissues and the express quantity of LHR in testis was the hightest. Testicular expression that quantity is epididymis 1.745 times, seminal vesicle 13.193 times, adrenal gland 50.505 times, brain 44.843 times, stomach 25.253 times, kidney 120.482 times. The LHR express quantity in tissues was significantly different, the distinction of which in testis and epididymis between autumn and winter was not notable; that in seminal, kidney, adrenal gland, brain, stomach was striking. The expression difference among tissues indicated that the affect of LHR to other physiological action was marked despite of reproduction. Also, the LHR expression of same tissues between autumn and winter is different. |
