| The hybrid of chicken-quail is a typical successful instance in poultry in hybridization.But the hatching rate of it is low,and the testis of it is dysplastic,and the hybrid of it is kakogenic, so it can not be used in production.Testis is the main reproductive organs and spermatogenesis places of male animal,but also it is a workplace where some certain hormone synthesis and secretion, it plays a key role on physiological and developmental. The testicular growth and development is affected by several factors. AR, ER genes may play a important role in testis development. The testis development is a cell proliferation and apoptosis process, and BCL-2 and PCNA is related genes which can effect the cell development and apoptosis. This study investigate the developmental expression patterns of AR,ER,PCNA and Bcl-2 genes in testis, using the method of Real time PCR. Meanwhile the morphological changes of testis were investigated by histol ogical method.The results are shown as follows:1. The morphological changes of testis in chicken,quail and the hybrid of chicken-quailApplication Anatomy and histology method on chickens, quail and the hybrid of chicken-quai of a comparative study of testicular morphology. Analysis the different testis in the method of anatomy and histology. Results:(1)The color of hybrid's testis is similar with the cheese's. The testis position and shape of chicken,quail and the hybrid of chicken-quail are different.It is smaller than chicken and quail.(2) The diameter of seminiferous tubules,the length of Seminiferous epithelium,and the volume density of seminiferous tubules are smaller than that of chicken and quail.In addition,the number of spermatogenic cell and sertoli cell is lower.2. Development patterns of testicular AR,ER mRNA expression in cock,quail and the hybrid of chicken-quail.(1) The expression of AR mRNA was increased then down in cock and quail. But a tendency of step-wise increase was observed in expression of AR mRNAchicken-quail.It suggests that AR genes has an affection on the development of chicken-quail's testis.(2) ER mRNA expression of testis in the hybrid of chicken-quail was significantly higher (P<0.01) from 10 to 50 days, and higher(P<0.05)from 50~90 days.And the expression patterns of them are different between each other,indicated that the expression of ER has species differences.3.Development patterns of testicular Bcl-2 mRNA expression in cock,quail and the hybrid of chicken-quail.The expression of Bcl-2 mRNA in Chicken testis increased gradually between 20 to 30 days, and decreased 50 days of age, then increased from 50 to 90 days gradually and reached a peak. It significantly decreasedat the age of 120 days (P<0.05) and get the lowest value. The expression of Bcl-2 mRNA of testis in quail decreased gradually and reached the bottom from 9 to 40 days, then rise rapidly and at a higher level. The expression of Bcl-2 mRNA of testis from 10 to 90 days unchanged on the whole. So we can see that hybrids did not has obvious cells apoptosis4. Development patterns of testicular PCNA mRNA expression in cock,quail and the hybrid of chicken-quail.The pattern of PCNA genes expression of testis in cocks and male quail were similar,which was declining at first, then rising significantly to the peak(P<0.01) but decreasing gradually, indicating that may play an important role in the development of testis in normal cocks and male quail. The expression of PCNA mRNA of testis in hybrid is not significant different, suggests that it may not be a obvious cell proliferation process in the development of testis in hybrid. |
