Viburnum opulus is belong to species of Caprifoliaceae Viburnum Linn, clusters of small deciduous shrub, which is one of very high development value of wild ornamental plants, and its flowering period is long, white flowers are fresh and elegant, the color of leaf changes with the season, rich small red fruit like cherries, attracting to the people a strong visual impact, the natural shape of stem and branches don't need pruning, reducing the landscape cost. Viburnum opulus L. also has medicinal value, plants contain chlorogenic acid, ursolic acid,β-sitosterol and flavonoids and other inqredients, Viburnum opulus L. is a plant species with medicinal uses of care for bone disease,cooling blood, invigorating the stomach and helping digesion, Swelling and relieve pain, for hypertension, loss of appetite, indigestion, arthritis, cough and other diseases. In recent years, with the development of urban greening, the market demand increasing for high ornamental value tree's number, species and the quality, but traditional breeding methods and natural variability on the way to breeding new varieties of low propagation coefficient, the efficiency of industrial production is not high, has been unable to meet the needs of production. Therefore the tissue culture virus-free technology for Viburnum opulus L. is a key process to solve this problem.This experiment takes tip and stems with buds as explants. the choice of explants, multiple shoot clumps induction, the proliferation of multiple shoot clumps, take root, and the process of transplanting various influencing factors asepsis, and Tissue culture seedlings and Original seedlings'physiological index were studied. The main contents focus on the following aspects, such as basic mediums and raising modes, the combination and density of hormone, different from the choice of virus-free technology and physiological index. The Orthogonal Design is adopted to arrange experiment. The results showed as following:1.Taking Viburnum opulus L. tip and stems with buds as explants, the best medium for: MS+6-BA2.0mgL-1+IBA0.1mgL-1+sucrose 20gL-1, the combination of germination rate of 95.0 percent.When different explants induced in the same medium, tip of the bud germination rate of 90 percent; followed by 2.0 cm long stems can reach 85 percent, with in a certain range the longer the explants, bud germination rate of the trend of higher. When inserting explants into the medium, the combination of germination rate is 85.0 percent. 2. The main factors of multiplication of bud clumps are 6-BA, NAA and sucrose. The results showed that:the most appropriate medium is MS+6-BA0.5mgL-1+NAA0.3mgL-+sucrose15gL-1, the best bud proliferation.3. Within the scope of the trial, in the medium 1/2MS+NAA0.2mgL-1+sucrose 15gL-1 the rooting results are the best.4. When transplanting, the first to go through one week of transitional period, then shade in the indoor ventilation, the average humidity of 70 percent to 80 percent of the greenhouse transplanting, vermiculite and perlite(1:1) survival rate was 85 percent, and the best seedling growth.5. To Viburnum opulus L. UV spectrum scanning. The results showed that:at 238nm and 352nm has absorption peaks. Measured indicators part of tissue culture seedling and original seedling, the results showed that:The total chlorogenic acid and chlorophyll content of the two indicators are more obvious physiological different, the tissue culture seedling is higher than the original seedling.
