Confocal Laser Scanning Microscope Developed

The Confocal Laser Scanning Microscope (CLSM) is a new kind of microscope that based on the Confocal Imaging Technology. Through the laser scanning and digital image manipulation,it could make the 3-D dynamic measurement of the sample. Compare with conventional microscope,it improved resolution and contrast of sample image. Now,it has wide application in biomedicine and material production-manufacturing fields.We develop a CLSM system included optics module,scan module,circuit and data collection module and computer processing module that can acquire reflected light and fluorescent light of sample. We debugged this system and measured some samples. Through the analysis of the measurement results,it is proved that the lateral resolution of our system is less than 1 micron.There are seven chapters in my thesis. In chapter one,the feature,common type and major applications of CLSM were introduced. In chapter two,the basic structure,operating principle and imaging character analysis of CLSM were introduced. During chapter three to chapter six,our CLSM system was introduced in details. In the last chapter,some matter in the debugging course,the measurement results of some samples using our CLSM and our prospect of CLSM system were introduced.
