The utilization of poultry farm waste is more and more widely recognized. However, the security of agricultural waste from the poultry farm was lack system study, especially the ecological characteristics of E. coli in poultry farm. Escherichia coli (E.coli) is widespreading in environments, which are prokaryotes and Gram-negative. Some of its specific serotypes can cause human and animal severe diarrhea and sepsis. E. coli can pollute water and soil by human and animal feces or by organic fertilizer and sewage, thus widely in the biosphere. As a representative type of environmental microbiology, Escherichia coli existence detection has been widely used in commercial organic fertilizer, organic fertilizer and environmental pollution. In bacteriological examination of environmental pollution, the coliform bacteria detection are indicators of fecal contamination and the number of coliform bacteria is determine the level of faecal pollution degree and the dangers sizes to human and poultry health. There study the ecological characteristics of E. coli and its affecting the safety of farm waste utilization.The main contents include: (1) study the number of E. coli presence and proliferation of dynamic in poultry farm; (2) analysis the popular serotypes of E. coli , 65 E. coli isolated from the typical colibacillosis lesions of illness and death poultry by positive serum for glass plate agglutination test; (3)study E. coli resistance spectrum and resistance patterns using antibiotic susceptibility test; (4)Survey the molecular epidemiology distribution of 14 types of virulence-related genes by PCR; (5) the ecological status of the bacteria are present by research avian Escherichia coli quorum sensing system. The results were followed:(1)E. coli is widespread in poultry farm environment, a range of E. coli peak on day 4 in per unit mass of organic manure, then decreased gradually; (2)Isolated 65 strains of pathogenic E. coli, in which O78 accounted for 75%, O2 accounted for 11% and O1 accounted for 5%, indicating that O78 is the major serotypes; (3)The phenomenon of multiple drug resistance widespread in isolates, 94% of the isolates resistance 10 types of antibiotics or drug, 100% of the isolates were resistant to rifampin and erythromycin, there is a wide range of resistance in avian pathogenic E. coli, veterinary drugs cause serious harm ;(4) ompA, pfs and luxs were conserved gene of E. coli in all 65 E. coli. The advantage of genotype of APEC is iss+iucD+tsh+cvaC+iroC+fimC+ompA+pfs+luxS+ genetic composition mode.(5)LuxS/AI-2 quorum sensing system is involved in regulating many physiological functions of bacteria, the luxS gene is involved in bacterial signal molecule AI-2 synthesis. To investigate whether there luxS/AI-2 type quorum sensing system in E. coli, V.harveyi BB170 bioassay demonstrated luxS functionality in E.coli, results showed that E.coli can produce AI-2 signal molecule .Further studies showed that AI-2 activity peaked in the late exponential phase.In summary, this study confirmed very unsafe the direct use of poultry farms waste and harmless waste treatment must be used in our current poultry production conditions by study the growth characteristics, resistance, serotype, virulence gene distribution and signal molecule AI-2 of E. coli in poultry market environment .
