Tripterine(celastrol)mainly presents in both Celastrus and Tripteryium, which has much pharmacological effects, such as anti-inflammation,antibacterial effect,immunosuppression,antifertility,etc. Recently, tripterine was found to be effective on antitumor. The purpose of this paper was to extract and isolate tripterine by advanced extraction and separation technologies, which was important for developing new medicines.1. Developing a HPLC method for determining the content of tripterine in the bast of Celastraceae, with this method 18 species of Celastraceae were determined. Tripterine was extracted in Soxhlet extractor by chloroform at 75℃, The HPLC determination was carried out on Kromasil C18 column (250x4.6mm,5μm) at 25℃, with the mobile phase of acetonitrile-water(80:20) which contained 0.005mol·L-1 H3PO4. The detection wavelength was set at 425 nm, and the flow rate was1.0 mL-min-1. Result show that the Celastraceae should be carried out to prepare tripterine.2. The extraction technique was studied. According to the property of tripterine in structure and character. In this process, four factors were discussed base on single factor experiments and orthogonal design. Then, the optimial extraction conditions of tripterine was achieved.3. The purification of tripterine was studied by column chromatography and deoiling,alcohol extracting-water precipitating the result were as follows:the later method had the characteristics of simple operation and high recovery, and good purification efficiency.It can servers as the pretreatment method. 4. The preparation of tripterine product was studied by the preparative liquid chromatography.The influences of the detection wavelength,the composition of mobile phase,the flow rate of mobile phase and the concentration of tripterine were investigated. Under the reasonable conditions,98.5%tripterine could be obtained.
