Objective:To investigate the forming artwork and to offer theory A/W for further research,we explored the difference about physico-chemical property and bacteriostatic action between two diverse drying artworks,drying under reduced pressure and spray drying,by which we prepared the "Qin Xiang Zhi Xie" dry extract.Methods:(1) To optimize the extraction artwork of costustroot by evaluating the weight of volatile oil;(2) To optimize such artwork as water-extracted,hypobaric drying and spray drying,orthogonal test was used to evaluate the influence of these factors,including the content of aesculin and aesculetin;(3) To explore the difference of physico-chemical property about dry extract between optimization of hypobaric drying and spray drying by evaluating such influence factors as appearance, hydroscopicity,flowability,thin-layer chromatogram,aesculin content and aesculetin content;(4) To investigate the difference of bacteriostatic action between the two kinds of dry extract described above by evaluating minimum inhibitory concentration and minimum bactericidal concentration for Escherichia coli,Bacillus dysenteriae,Staphylococcus aureus and Bacterium vulgare.Results:The best condition about extraction costustroot volatile oil is follows:recirculation for 4 h;water-extracted for 2 times with the volum of water 8 times or 6 times of medical material.and recirculation for 1.5 h or 1 h respectively.The ideal temperature of hypobaric drying is 70℃.By spray drying,the concentrated solution density is from 1.14 to 1.16 and the temperature of the wind is 160℃.Extract under spray drying charactered that color uniformity,not easy moisture absorption,good flowability and clear spotting in the lamellar icon;Nevertheless,by hypobaric drying the content of aesculin is higher and better bacteriostatic action.Conclusions:Weighing the results of physico-chemical property and bacteriostatic action;fungistasis,we arrived at the conclusion that hypobaric drying has more advantages than spray drying.
