| Agaricus blazei Murill is a valuable food-medicine fungi. In recent years it was found that Agaricus blazei has a wide range of biological activity which was closely related to its polysaccharide content.Orthogonal design and uniform design was used in the experiment to study the best conditions of immobilization of the three enzymes and their infact on polysaccharide extracts rate of Agaricus blazei,the conclusions are as follows :The most rational technological factors for extraction of Agaricus blazei Murill polysaccharide with compound enzymes under ultrasonics wave were studied in this paper. Uniform design was used to analysis the influence of different conditions including times, pH values, temperatures, the amount of enzymes and stock-extract ratio on enzymatic process of cellulase, pectolase and papain, and influence of two different ways of addition of enzymes on polysaccharide extracting rate and the best condition of ultrasonic wave complex enzymatic technology were studied. The results showed that the extracting rate could increased by ultrasonic wave complex enzymatic technology. The best technological conditions for extraction were as follows: first add pectolase (the amount was 7000U/g ,the ratio of material and water was 1:45 ,pH value was 3.8, extracting for 90 min at 50℃), then cellulase (the amount was 150U/g ,the ratio of material and water was 1:45 ,pH value was 3.86,extracting for 120 min at 75℃),finally papain(the amount was 20000U/g ,the ratio of material and water was 1:45 ,pH value was 3.6,extracting for 120min at 75℃). The extracting time was 20 min., the yield of polysaccharides is up to 14.51%.The results showed that the use of immobilized compound enzyme extract Agaricus blazei Murill got the highest yield of the polysaccharide , the best technological condition for immobilization and extraction are as follows: The best conditions for immobilized compound enzymes are as follows:the concentration of alginate and gelatin was 1.0% and 3.0% respectively, the radio of cellulose ,papain and pectolase was 1:1:1.4,pH value was 5.0 and the temperature was 50℃.In this condition the activity recovery of each enzymes was 52.74%,52.98% and 56.93 seperatly.The best extraction conditions of immobilized compound enzyme are as follows: solid-liquid ratiol:35;the volume of Agaricus:immobilized enzyme was 10:1.28 (that is, adding considerable cellulase 15 U/g, pectinase 293 U/g, papain 5000 U/g); enzymolysis time for 2.0h;then extraction by hot water for 2.5h. the average yield of polysaccharide was 18.65%. |
