The plant of aloe belongs to Liliaceae, it is an important medicinal origin plant, which is well known to have pharmacological activities such as diuretic, diarrhea and anti-hypertension that have been conventionally used as healthy beverage with many people from ancient times. Modern research showed anthraquinone has multi-functions including antibacterial, antioxidant and antimutagen, and be wildly used in medicine, health care products, food additives. The craft conditions of the extraction, purification and isolation of anthraquinones were studied. For better use of aloe, it is important to study its extraction and purification, find the optimum craft conditions.According to spectral characteristics of anthraquinone, the ultraviolet -visible spectrophotometry was established to determine the content of anthraquinone in aloe. According to the results of stability experiment, reproducibility and recovery experiment, proved that it had good stability, high reproducibility, recovery, and easy to operate, can be use in production process.The extraction conditions of anthraquinone with ethanol extraction were carried out. The influence to extraction factors such as ethanol concentration, extracted temperature, extracted volume, extracted time were investigated. By uniform design experiment, we confirmed the optimum conditions was 60% ethanol, the ratio of solid to liquid was 1:10, extracted time was 6 hours, extracted temperature was 30℃.Secondly, the hydrolysis conditions of anthraquinone(glycoside) was carried out. The influence to hydrolyzation such as sulfuric acid concentration, liquid-solid rate, hydrolysis temperature, hydrolysis time, mix round speed were investigated. by uniform design experiment, we confirmed the optimal sulfuric acid concentration was 2.0mol/L, liquid-solid rate was 1:10, the reaction temperature was 130℃, the reaction time was 3.5h, the mix round speed was 10rps.Lastly, for isolation of anthraquinones, the adsorption and desorption performance of 3 kinds of macroporous adsorbent resins was studied and compared, and polyamide resin was selected for its excellent adsorption and desorption properties. The effect of concentration, pH and work speed of the infusion on the adsorption of the polyamide resin was studied with dynamic adsorption experiment. The follow rate was 1.0mL/s, solution concentration was 0.22mg/mL and the pH was 5; After eluted with 2BV of distilled water and 4BV of 80% ethanol, the yield of anthraquinones was 91.1%, and product purity was 49.2%. We did adsorption-desorption and regeneration experiment of Polyamide Resin, we finded adsorptive capacity and elution efficiency of Polyamide Resin was reduced after 6 times, we must to regeneration. After 4 times, the Polyamide Resin can't be use.
