The experiment assessed the toxicity of effluent from sewage treatment plant of north city applying the early embryo of Japanese medaka (Oryzias latipes) from morphological and molecular level. The test showed that it appeared the phenomena of embryo lesions, falling of hatch ratio and rising of deformities ratio four groups including 4000%,100%,50% and 25% concentrations. Embryo lesions include the spawn membrane rupturing and embryo cruor mainly and deformities consist of inhibiting development, small eyes, abnormal eyes, notochord curling, and yolk sac shrinking, so it show effluent still having the toxicity. To find the matter resulting in development deformities of the early embryo, through three chemical of 17β-Estradiol,indirubin and retinoic acid exposure experiments,it find out that the retinoic acid can induce deformities of embryo of Japanese medaka about 10ng/L,but 17β-Estradiol and indirubin can not. Using three chemical together exposure experiments, 17β-Estradiol and indirubin can enhance the toxicity effect of the retinoic acid. With the ingredient having RA activity of sewage exposure experiments, it discover the ingredient have the similar deformities effect with the retinoic acid, and make the Comparison with it from gene expression level. Therefore, it judge the ingredient of similar with retinoic acid is main matter of resulting in development deformities of the early embryo.
