At present, domestic exploitation success natural tocopherol preparation technology could well adequate for deodorization distillate of high tocopherol content (more than 5%),but limited by technology and equipment, the content of deodorization distillate under 2% in existence domestic, which were hard to make use of. The proportion of liquor and raw material was 4.5 L/kg, alcohol pH was 4.0,92% alcohol extraction temperature was 64℃, mechanical agitation rate was 120 r/min, backflow extract 20 min.The extraction rate was 68.8 %, and the degree of fineness was 1.81 %. Single factor experiment optimum elution condition: silica gel 32.5g ,ethanol-petroleum ether mixed liquor, elution solvent blending ratio was 20: 80, elution velocity 1.2 mL/min, ratio of high to diameter was 10, sample loads was 3.6g,solvent dosage was 240 mL. From 100th to 170thmL eluent was Collected, natural tocopherol concentrate 3.12 times after removal solvent, recovery rate was 81.2%, the content was 5.65%. Silica gel regeneration,elution solvent were reclaim nest. The content of natural tocopherol was satisfactory to the content of subsistent natural tocopherol preparation technology.Colza oil deodorization distillate was aqueous phase saponified, the saponification technology was optimized by planning, the proportion of sodium hydroxide and rough material was 0.2kg/kg, the proportion of water and rough material was 5L/kg, backflow saponified 8h under 80℃, unsaponifiable matter was gained after saponification solution was washed by saturation saline solution.Extraction technology of phytosterol from unsaponifiable matter by ethyl acetate was optimized by response surface method, the proportion of liquor and raw material was 7 Lethyl acetate per kilogram deodorization distillate, extraction temperature was 62℃,mechanical agitation rate was 140 r/min, back flow extract 40 min, 1000 r/min centrifuge 15 min, phytosterol crude product purity was 15.18% after reclaim solvent from liquid extract, the extraction rate was 75.97%. Phytosterol concentrate effect was remarkably which analyzed by the HPLC. |
