| Lingdan tablets were consisted of Ganoderma, Salvia miltorrhize (SM) and Ligusticum Chuangxiong ,which had been applied in clinical therapy at Wuhan NO.1 Hospital for many years. As a therapeutic measure to different kinds of lesiondil follicles, it was very effective to mourish Yin and tonify the kidney ,dissolve stasis on blood lipid and hemorheology, improve hair-growth. So Lingdan tablets had been generally welcomed by clinical doctors and patients. The old preparation was sugar-coated tablets, which had a lot of defects and was lack of content determinations. Based on sugar-coated tablets, orthogonal design experimentions was used to study the precedure, HPLC was used to determine tanshinone Ⅱa of the tablets, the dose style was designed as film-coated tablets. To establish qualitative and quantitative analysis of lingdan tablets, the study set a high priority on the extraction methods and preparation of ganoderma and SM. All these provided the scientific foundation for rational industrial production. Furthermore, it has a marked clinical therapeutic effection on alopeia arenta (AA) and male pattern baldness (MA).TLC and chemical identify were used to qualitatively analyze the components of the tablets, while HPLC was used to determine tanshinone ⅡA and danshensu. Lichrosorb-C18(46mm×150mm,ID=5 μm) was used as the chromatographic column. Methanol-water (75:25) was taken as the mobile phase and detection walvelength at 270nm. The contents of polysaccharides in Lingzhi was detected at 625nm by visible-spectrophometry. The best extraction and preparation precedure were optimized by orthogonal design experimentions.From the results, we could conclude that these qualitative and quantitative analysis of lingdan tablets were convenient, sensitive and accurate.
