| The fruit of Polygonum hydropiper L., one of the genus Polygonum, contains abundant flavonoids and macro elements K and Ca. Flavonoids has many kinds of important physiological functions and an extensive application prospect in the fields of medicine, cosmetics and health products. Macro elements K and Ca are essential elements for the human body and mean a great deal to the life. This paper that was concerned with the Flavonoids and macro elements K and Ca from the fruit of Polygonum hydropiper L. systematically studied the technologies of the extraction and purification of the total flavonoids, determined the distribution situation of K and Ca at the same time and would offer the experiment basis for further developing and utilizing the fruit of Polygonum hydropiper L. The following researches have been carried out:1. The analysis of the total flavonoids The qualitative analysis of flavonoids by colouring showed that the extraction solution contains flavonoids. The content of the total flavonoids from the fruit of Polygonum hydropiper L. was analyzed quantitatively by spectrophotometry and effect of parameters including color time, degree of precision, repeatability, recovery of the samples on measuring result was investigated. The optimized measuring condition was as follows: wavelength 510nm, color-developing agent 5%NaNO2-10%Al (NO3)3-4%NaOH.2. The extraction of the total flavonoids The extraction process of the total flavonoids from the fruit of Polygonum hydropiper L. by single factor and orthogonal test was studied. The optimum extraction condition was established as follows: solvent 70% (v/v) ethanol, extraction time 2.0h, extraction temperature(water bath) 85℃, extraction times twice and ratio of material to liquid (g/mL) 1 I 15. Under these conditions, percentage composition of the total flavonoids was 6.027%, and the abstraction rate of K and Ca was 3.028% and 0.229% respectively.3. The purification of the total flavonoids While the total flavonoids was purified by liquid-liquid extraction, the percentage loss of the total flavonoids was 27.26%, the turbidity of the liquor purified was 7.50, percentage purity was 70.88%, and the yield of K and Ca was 82.734% and 95.570% respectively. While the total flavonoids was purified by member method, the percentage loss of the total flavonoids was 0, the turbidity of the liquor purified was 6.50, percentage purity was 46.59%, and the yield of K and Ca was 100% and 70.973% respectively. Furthermore,the optimum purification technology of the total flavonoids by 0.1 u m filter membrane — macroporous adsorption resin method was determined, namely the contact time of adsorption 0.35h, the concentration of under-adsorpted solution l.Omg/mL, pH 3.5, under-adsorpted solution .' resin(mL/g)=7 ." 1, eluant ethanol, the contact time of desorption l.Oh, the concentration of the eluant (v/v) 85%, eluant '. resin(mL/g)=7 '. 1. Under these conditions, the percentage loss of the total flavonoids was 39.76%, the turbidity of the liquor purified was 4.0, percentage purity was 50.17%, and the yield of K and Ca was 27.049% and 25.154% respectively.Comparing the three sorts of purification methods, we could conclude that purify the total flavonoids from the fruit of Polygonum hydropiper L. by 0.1 u m filter membrane—macroporous adsorption resin should be a promising technology. |
