The chromatogram of reversed phase high performance liquid chromatographic fingerprints of the four kinds of Epimedium L. crude drugs was established. Five kinds of the compound in the extract were separated and prepared by high performance reversed phase liquid chromatography (RPLC), and their structures were investigated respectively. This thesis includes five parts as follows:1. ReviewA review of the most components in Herb Epimedii and the quality standard of Chinese traditional medicine were given. It contains ninety six references.2. Extraction of effective components from Herb EpimediiThe extration efficiencies of icariin from Herb Epimedii by aqueous solution of ethyl alcohol, acetic acid were investigated and compared with each other. The extraction efficiencies of icariin by soaking at room temperature, refluxing and ultrasonic method were also compared. Extraction mode and extraction time were confirmed on this basis. The result shows that the effective components of Herb Epimedii can be extracted by 70% ethyl alcohol aqueous solution by ultrasonic extracted with thirty minutes.3. Separation of effective components from Herb Epimedii Optimization of the separation condition of Epimedium Sagittatum extract wasseparated by a multi-step gradient elution method by RPLC. Using acetonitrile aqueous solution as mobile phase, the Epimedium Sagittatum extract was separated well and nineteen components with good separation were abtained. Under the optimized gradient conditions, the retention time and resolution of Epimedium Sagittatum extract show a good stability and reproducibility. Using the method described in this paper, the analysis time of Epimedium Sagittatum extract was shortened and good separation.4. Studies on the RPLC fingerprints and quality evaluation of Epimedium L. crude drugsWith a new multi-step gradient elution, the chromatograms of reversed phasehigh performance liquid chromatographic fingerprints of the four kinds of Epimediurfi L. crude drugs producing in Qinling Mountainous area were firstly established. The common RPLC fingerprint of the Epimedium L. crude drugs containing nine components including icariin was found. The icariin was selected as the internal standard and in terms of the relative amount of the nine components in the RPLC fingerprints of the four kinds of Epimedium L., the quality of the crude drugs was also evaluated. However, because of different producing areas and subgenus, each of the four RPLC fingerprints has specific character itself. In addition, it was also found that the fingerprint of different producing areas of the Epimedium sagittatumm Maxim consist of twelve components. The RSD for the precision, stability and repeatability of all measurement are less than 3%. This method is simple, rapid, stable and has a good reproducibility. It may be used to identify the kind and to evaluate the quality of Epimedium L. crude drugs, and also to provide the scientific evidence for exploring some of available component of this drugs in the future. 5. Preparation and structure determination of five compounds fromthe Herb EpimediiIsolation and preparation of five main compounds from the extract of Epimedium Sagittatum by RPLC were studied. Structures of the five compounds were identified by UV, IR, MS, 'HNMR ,i3CNMR and MALDI-TOF-MS.
