Studies on separation of protein and polypeptide using chip based capillary electrophoresis system were performed in this work.In the first section, the application of microfluidic chips on separation of protein and polypeptide was reviewed, including the chip materials, detection techniques, separation modes and the surface modification methods for microchannel wall.In the second section, a chip based CE system with laser induced fluorescence detector was developed to achieve the separation of protein and polypeptide. Bovine serum albumin and trypsin inhibitor are separated with this system. The main work was focused on the separation of fluorescein isothiocyanate labeled tryptic digests of bovine serum albumin. The pinch injection mode was employed for sample introduction. The effects of buffer composition and separation channel length on the performance were investigated. Under the optimized conditions, the separation of polypeptide sample was achieved within 250 s.In the third section, based on the established hybrid microfluidic system coupled a glass chip with capillary to achieve UV absorbance detection, the separation of the mixture of paracetamol, sulfamethoxazole, trimethoprim and theophyline was realized.
